Section 3.1 - Reagents for Modifying Alcohols

چکیده
Although alcohols (including phenols such as tyrosine and the alcohol groups in serine, threonine, sterols and carbohydrates) are abundant in biomolecules, their chemical reactivity in aqueous solution is extremely low. Few reagents are selective for alcohols in aqueous solution, especially in the presence of more reactive nucleophiles such as thiols and amines. It is therefore difficult to selectively modify serine, threonine and tyrosine residues in proteins except when they exhibit unusual reactivity, such as by residing at an enzyme’s active site.

Catalytic asymmetric alkylation of ketones using organometallic reagents.
The catalytic asymmetric synthesis of tertiary alcohols by the addition of organometallic reagents to ketones is of central importance in organic chemistry. The resulting quaternary stereocentres are difficult to prepare selectively by other means despite their widespread occurrence in natural products and pharmaceuticals. Over the past few years, several seminal reports on the formation of chi...
متن کاملEfficient and chemoselective alkylation of amines/amino acids using alcohols as alkylating reagents under mild conditions.
We report a mild and environmentally benign method for the synthesis of tertiary amines using alcohols as the alkylating reagents. Not only secondary amines such as piperazines but also amino acids and amino alcohols can be N-alkylated selectively. For N,O-benzyl protected amino alcohols, both N,O-de-benzylation and N-methylation were achieved in one-pot.
متن کاملUniversity of Groningen Synthesis of Optically Active beta- or gamma-Alkyl-Substituted Alcohols through Copper-Catalyzed Asymmetric Allylic Alkylation with Organolithium Reagents
An efficient one-pot synthesis of optically active β-alkyl-substituted alcohols through a tandem coppercatalyzed asymmetric allylic alkylation (AAA) with organolithium reagents and reductive ozonolysis is presented. Furthermore, hydroboration−oxidation following the Cucatalyzed AAA leads to the corresponding homochiral γalkyl-substituted alcohols.
متن کاملSynthesis of highly substituted allylic alcohols by a regio- and stereo-defined CuCl-mediated carbometallation reaction of 3-aryl-substituted secondary propargylic alcohols with Grignard reagents.
A highly regio- and stereoselective CuCl-mediated carbometallation reaction of 3-aryl-substituted secondary propargylic alcohols with alkyl, aryl, vinyl or allyl Grignard reagents for the synthesis of fully-substituted allylic alcohols was developed. The R(2) group from the Grignard reagent was successfully introduced to the 2-position of the propargylic alcohols due to the chelation of metal a...
متن کاملStereoselective Synthesis of Steroids and Related Compounds, IV['l Addition of Cerium Reagents Derived from (Trimethylsily1)propargyl Bromide to Aldehydes
The addition of (trimethylsily1)propargylmagnesium bromide (9) to aldehydes afforded predominantly the allenylic alcohols rac-10. The regioselectivity of this addition changes dramatically when organocerium reagents were used for the addition. In this case nearly exclusive formation of the homopropargylic alcohols rac-8 was observed. Highest selectivity for this process was obtained when cerium...
